During testing, we established that the columns retain heat after FISH incubation for 10 min at 80 °C. The plate should be immediately cooled at 4 °C to prevent slow reannealing of DNA and to improve signal to noise ratio. More stringent post-FISH wash with lower SSC buffer concentration should be performed to reduce cytoplasmic background staining.   Modifications: Users may attach glass coverslips or aluminium foil to the base of each column to create a flatter surface and promote ease of cleaning.
